Isolating peripheral blood mononuclear cells (PBMCs)

I want to share with you a little bit of what I went through during the  3 weeks training in Karachi. This is a brief article on how isolation of peripheral blood mononuclear cells was carried out. Blood from healthy donor, mixed with media, then layered on gradient-density medium. After that, it will be centrifuged and it will give us layers of cells in the tube. We will take the the layers containing PBMCs.


In the video below, it showing blood is mixed with media, before we layer it on the gradient-density medium.

 

 

The gradient-density medium sometime branded as Ficoll, other supplier will name it Lympho Separation Medium or Lymphoprep.

The result, the gradient-density medium will separate the blood to several layers through the centrifuge force. The layers containing certain group of cells, based on their weight / density. Sorry for the time being, I don't have any picture or video to show how the layers look like.

 

There are still a lot more steps in the experiment. Will publish it later.
